The wide availability of inexpensive synthetic single-stranded DNA and RNA has opened the way for routine DNA amplification by PCR. This use of oligonucleotides as primers has revolutionized the study of gene expression and disease processes. They are most commonly used as antisense oligonucleotides, small interfering RNA, primers for DNA sequencing and amplification, probes for detecting complementary DNA or RNA via molecular hybridization, tools for the targeted introduction of mutations and restriction sites, and for the synthesis of artificial genes. In a type of DNA sequencing known as dideoxy sequencing, oligonucleotides are used as a primer, so that the enzyme that makes the DNA will have a template to work from. Single-stranded DNA is used, and an oligonucleotide that is complementary to the DNA strand is synthesized using an automated machine. Note: At 100 µM concentration, 1 µL solution contains 100 pmol of oligo.
